…Tour Ste. Saulte Marie area. Visit Agawa Canyon. Visit Saulte locks.

SSM is okay but it's the natural sites near there and on the route to Toronto that are worthy of your time. Hug the shore of the North Channel and Georgian Bay and be open to getting out of the car to take in some of the great things on the way.
Can one view the July 4th fireworks over Niagara Falls from Toronto?
Nope, not a chance. It's 43 miles away as the crow flies.
Is one full day in Niagara Falls enough?
Yes, unless you like casinos. Take the time to visit NF's parks near the Falls. And drive the Niagara Parkway and visit the lovely town of Niagara-On-The-Lake.
Is two full days (and 3 nights) in Ste. Saulte Marie too much?
IMHO, yes.
Is two full days (and 3 nights) in Toronto too much/not enough?
It's definitely enough to give you a good flavor of the place.
Anything amazing on the stretch of the Trans-Canada highway between the two?
If you get a chance to do some kind of boat tour in the 30,000 Islands area of Georgian Bay, do it.

If you want to see what multi-million dollar cottages look like, noodle around Lake Joseph, Lake Rosseau, and Lake Muskoka in Muskoka.
