I upgraded from a 24" iMac to a 27" iMac and have not regretted it once. In order of importance the three factors that got me to spend the money on a newer computer were:
1) size of monitor
2) size of internal hard drive
3) speed
Sold the 24" iMac and kept the wired keyboard with numeric keypad. (Working in the back office of a bank during college taught me to touch type with a keypad.)
Have got a Magic Mouse, an Evoluent 3 vertical wired mouse, and a trackpad on the desk. The Evoluent and Magic Mouse live to the right of the keyboard, the trackpad to the left.
While I really like the Magic Mouse a lot, I reach for the Evoluent vertical mouse much more frequently. Next in frequency of use is the trackpad. I think that's mostly to switch hands, rather than the design of the trackpad, though it's nice.
Every so often I use the Magic Mouse. When I do, I'm reminded how nice it is, and then go back to the Evoluent mouse. The vertical aspect just feels more natural and comfortable to my hand and forearm.
