yes if it's configured to be 'compatible'. Original card is 802.11b (10 MBps). Airport Express is 802.11g (54MBps) and backward compatible / capable with 802.11b if and only if it is configured that way. The old grey Airport Base Station is 802.11b.
The new Airport Extreme base stations are 802.11n (100MBps), and are backward compatible with 802.11g and 802.11b if and only if it is configured that way.

Easiest way to see if your card on your current laptop is working is to look for other wireless access points (assuming you've got Airport port enabled on your Locations preferences).
